Construct scFv-Fc of the starting IgG
Establish the base line antigen binding using scFv-Fc and surface displayed scFv
Design and construct focused libraries and screen for higher binding affinity
Isolate top 10 scFv clones, and measure binding affinity in comparison to starting scFv
Recover DNA sequences of confirmed clones
Reconstruct scFv-Fc and purify 100 ug protein
Measure binding affinity of purified scFv-Fc using ELISA or Biacore in comparison to starting scFv
