Description:
Monoclonal antibodies to clinically important target proteins have been developed and used as therapies to a wide variety of diseases in humans. Monoclonal antibodies are usually produced in non-human species, most often in mice. However, to become successful biotherapeutics, these non-human antibodies have to overcome several significant drawbacks. First, non-humanized antibodies can elicit an immunogenic response when administered to patients, as the human immune system can recognize the non-humanized antibodies as foreign. Additionally, non-humanized antibodies frequently have a short in vivo half life in humans. Creation of chimeric antibodies (non-human variable regions with human constant regions) in some situations can partially alleviate these problems. However, a much more effective strategy is the complete humanization of the antibody, which involves transferring the antibody CDRs into a proper human antibody framework, while retaining its original affinity and specificity.
LakePharma has developed and validated a platform for antibody humanization that combines the advantages of both rational and empirical approaches. The antibody variable sequence is queried against an annotated human antibody database, and three distinct humanized light chain and three heavy chain sequences are selected based on LakePharma’s proprietary primary sequence homology and 3D structure modeling programs. All nine combinations of designed light and heavy chains are constructed and then produced recombinantly in CHO cells. All nine are tested for production levels as well as affinity to the original antigen. The top three combinations will be produced at high levels in CHO cells and delivered to the client as 100 ug purified antibodies. Based upon the customer’s needs, LakePharma can perform additional affinity maturation on the humanized antibodies to identify variants with even greater binding affinity.
